Background The upregulation of interferon (IFN)-stimulated genes (ISGs) induced by type I IFNs (namely IFN signature) in RA patients had implications early diagnosis and prediction therapy responses. However, factors that modulate the signature are largely unknown. Objectives To explore involvement VGLL3, a homologue vestigial-like gene Drosophila putative regulator Hippo pathway, modulation RA. Methods FLS were isolated from osteoarthritis patients. Expression VGLL3 synovial tissues was analyzed immunohistochemistry PCR. RNA sequencing performed RA-FLS upon overexpression. expression ISGs examined PCR Western blotting. Results upregulated synovium compared to OA. Overexpression promoted RA-FLS. STAT1 MX1 also associated with OA IRF3 activation, IFN-β1 expression, autocrine signaling modulated pathway molecules WWTR1 AMOTL2, which mediated regulation activation production Conclusion drives IRF3-induced secretion inhibiting subsequently promotes through signaling.
